RESUMO

OBJECTIVE@#To investigate the effect of zoledronate (ZOL) on osteoclast differentiation and bone resorption under high glucose, and the regulation mechanism of p38 mitogen activated kinase (p38 MAPK) signaling pathway in this process.@*METHODS@#RAW264.7 cells were divided into four groups: low group, high group, low+ZOL group and high+ZOL group after induced into osteoclasts. Cell proliferation activity was determined by MTT assay. The migration of RAW264.7 cells were examined Optical microscopy. Immunofluorescence microscopy was used to observe the cytoskeleton and sealing zones of osteoclasts. After adding group 5: high + ZOL + SB203580 group, trap staining was used to identify the number of positive osteoclasts in each group. The number and area of resorption lacunae were observed by SEM. The mRNA and protein expression of osteoclast related factors were detected by real-time PCR and Western blotting.@*RESULTS@#The cells in the 5 groups showed similar proliferative activity. High glucose promoted the migration of RAW264.7 cells (@*CONCLUSIONS@#High glucose inhibits osteoclast differentiation and bone resorption. ZOL inhibits osteoclast differentiation and bone resorption in high-glucose conditions by regulating p38 MAPK pathway, which can be a new pathway for ZOL to regulate diabetic osteoporosis.

RESUMO

Objective To investigate whether 5% strontium-containing brushite bone cements(DCPD) has repair effect on alveolar bone defects in osteoporotic rabbits. Methods Eighteen healthy adult female rabbits were used to establish osteoporosis models and were randomly divided into three groups:the blank control group, DCPD group,doped 5%DCPD group(5%strontium),with 6 rats in each group.In addition to the blank group, rats the other groups were filled with the corresponding bone cements in the bilateral alveolar bone defects. At 4 weeks and 8 weeks after operation,3 rats in each group were killed and given shooting the defect area X-ray.The expression of b-FGF was detected by immunohistochemistry. Results X-ray results showed that the defected of doped 5% DCPD group have been nearly completedat the 8 week,DCPD group partially completed repair,but blank group was not fully repaired. The results of immunohistochemistry showed that the expression of b-FGF was the highest at 4 weeks and decreased at 8 weeks after operation.Expression of b-FGF was significantly different at 4 weeks and 8 weeks among the three groups.Conclusion The 5%strontium-containing brushite bone cements can repair bone defect in osteoporotic rabbits.

RESUMO

Objective:To study the effects of brushite coating with strontium on the osteoblast-related factor expression of MC3T3-E1 cells.Methods:Brushite coating on tinanium surface was prepared by electrochemical deposition and the extract of the coating with stronium at 0.1%,0.5% and 1% were prepared respectively.MC3T3-E1 cells were cultured with the extracts,cell prolifera-tion,ALP activity,bFGF and VEGF mRNA were examined by MTT assay,ALP staining and RT-PCR respectively.Results:Stronti-um containing extracts promoted the proliferation,increased the ALP activity and mRNA expression of bFGF and VEGF(P <0.05)of MC3T3 cells.Conclusion:Brushite extract containing strontium may promote osteogenic function of MC3T3-E1 cells.

RESUMO

Objective To summary our experiences in percutaneous nephrolithotomy for treating the caliceal diverticular calculi and postoperative outcome with technical modification.Methods A retrospective review was performed on 34 patients with caliceal diverticular calculi who underwent percutaneous nephrolithotomy between January 2009 and June 2013.The patients were divided into 2 groups.In group A (1 1patients),stones were removed and diverticular neck was incised.In group B (23 patients),fulguration of diverticular wall was performed after the stone removal and diverticular neck incision.Group A had 11 cases including 4 male and 7 female with average age (36.7± 16.0) years.Mean size of stone was (1.8±0.4) cm.Mean maximum diameter of diverticulum was (2.3±0.5) cm.Group B had 23 cases including 10 male and 13 female with average age (40.1±12.0) years.Mean size of stone was (1.9±0.6) cm.Mean maximum diameter of diverticulum was (2.3±0.8) cm.There were no significant difference in stone size and maximum diameter of diverticulum within two groups (P>0.05).The following parameters such as operative time,drop in hemoglobin,stone clearance,complication rate and ablation rate of diverticulum were compared between two groups.Results In group A,average operative time was (62.7±11.7) min.Mean hemoglobin dropping was (12.0t9.7) g/L.Stone clearance rate was 90.9% (10/11) and overall complications rate was 18.2% (2/11).The ablation rate of diverticulum was 63.6% (7/11).In group B,average operative time was (76.3±21.6) min.Mean Hemoglobin reducing was (12.9±16.7) g/L.Stone clearance rate was 91.3%(21/23) and overall complication rate was 21.7%.The ablation rate of diverticulum was 91.3% (21/23).There was no significant difference in stone clearance and complication rate between two groups.It seemed that the operativc timc in group B was longer than that in group A.However,no significant difference was found (P>0.05).The ablation rate of diverticulum in group B was significantly higher than that in group A (P<0.05).Conclusion In percutaneous nephrolithotomy treatment for caliceal diverticular calculi,fulguration to diverticular wall is an effective method to achieve diverticular obliteration and reduce possibility of stone recurrence.

BACKGROUND:Tartrate-resistant acid phosphatase is a specific marker for osteoclast differentiation and bone resorption, which is a sign of osteoclast maturity. OBJECTIVE:To study the effect of alendronate on tartrate-resistant acid phosphatase related to osteoclast differentiation and bone resorption. METHODOsteoclasts were cultured by mouse monocyte-macrophage cellline-RAW264.7. The cells were divided into two groupcontrol group, treated with 100μg/L receptor activator of nuclear factorκB ligand factor;alendronate group, treated with 100μg/L receptor activator of nuclear factorκB ligand factor+10-7 mol/L alendronate. Osteoclastogenesis and resorption function of osteoclasts were examined at 7 days of culture. Gene expression of tartrate-resistant acid phosphatase was detected by immunofluorescence method. Western blot assay was used to detect protein expression of tartrate-resistant acid phosphatase. RESULTS AND CONCLUSION:Tartrate-resistant acid phosphatase positive multinuclear cells were observed and resorption lacunae formed in two groups. Control group showed the higher number of tartrate-resistant acid phosphatase positive multinuclear cells and larger size of resorption lacunae than the alendronate group (P<0.01). Immunofluorescence showed expression of tartrate-resistant acid phosphatase was higher in the control group than the alendronate group (P<0.01);furthermore, the protein expression of tartrate-resistant acid phosphatase was also lower in the alendronate group than the control group (P<0.01). These findings indicate that bisphosphonates could strongly inhibit osteoclastogenesis and its resorption function by inhibiting protein expression of tartrate-resistant acid phosphatase.

BACKGROUND:Studies have shown that bisphosphonates inhibit osteoclast resorption, but whether cathepsin K, a key cytokine of bone resorption, plays an effect has rarely been reported. OBJECTIVE:To study the effect of bisphosphonate on capthesin K and bone resorption function during osteoclast differentiation. METHODS:Osteoclasts were cultured by mouse monocyte-macrophage cellline-RAW264.7. The cells were divided into two groups:control group, treated with 100μg/L receptor activator of nuclear factorκB ligand factor;alendronate group, treated with 100μg/L receptor activator of nuclear factorκB ligand factor+10-7 mol/L alendronate. Osteoclastogenesis and resorption function of osteoclasts were examined at 7 days of culture and gene expression of capthesin K was detected by immunofluorescence method at 72 hours of culture. Western blot assay was used to detect capthesin K protein expression at 72 hours of culture. RESULTS AND CONCLUSION:Tartrate-resistant acid phosphatase positive multinuclear cells were observed and resorption lacunae formed in two groups. Control group showed the higher number of tartrate-resistant acid phosphatase positive multinuclear cells and larger size of resorption lacunae than the alendronate group (P<0.01). Immunofluorescence showed expression of capthesin K was higher in the control group than the alendronate group (P<0.01);furthermore, the protein expression of capthesin K was also lower in the alendronate group than the control group (P<0.01). These findings indicate that bisphosphonates could strongly inhibit osteoclastogenesis and its resorption function by inhibiting gene expression of capthesin K.

RESUMO

Objective To discuss the Survivin protein expression in different salivary gland tumor.(Methods)This experiment was based on tissue chip technology.Expression of Survivin protein in the salivary gland tumor was analyzed with immunohistochemical methods.Results ① Survivin expression showed significant difference in the salivary gland benign and malignant tumors(P0.05).The gland around the malignant tumor showed diversity compared with malignant tumor(P0.05).Conclusion Survivin expression has correlation with the relay tumor canceration,and the gland around malignant tumor has correlation with occurrence,progress,prognosis in the salivary gland malignant tumor,and it is a reliable marker of earlier period diagnosis and prognosis of the malignant tumor of salivary gland.
